ParoF--mCherry--dam
Dam is involved in methylating DNA, overexpression causes 6.4 * 10^-4 /// 6.4 * 10^4 mutation rate increase, mostly catalyzing transversions. It requires no mutations and simple overexpression is sufficient to cause its mutagenic activity. In this plasmid, it is placed downstream of the tyrosine-repressible promoter ParoF and reporter gene mCherry to fulfill our goals to improving an established system of directed evolution.
